Clarity OTX extraction kits from Phenomenex Inc., Torrance, Calif., speed up the isolation of RNA and DNA-based therapeutics and their metabolites from ADME/ pharmacokinetics studies in preparation for downstream LC/MS analysis. While traditional methods require eight steps and can take two hours, the Clarity OTX extraction kits isolate oligonucleotide therapeutics and their metabolites in 15 minutes. It can be used for both plasma and tissue; other methods require separate procedures for each sample type.
The solutions combine a solid phase extraction (SPE) sorbent with buffers to deliver high recovery of all types of oligo therapeutics. The kit solution can be scaled up to a 96-well plate format for automated or parallel processing for high-throughput environments.
The Clarity OTX protocol uses a propriety mixture of chaotropes, detergents, and other dispersants to disrupt intermolecular interactions that are common for oligonucleotides in biological matrices. A proprietary mixed-mode sorbent captures and retains small synthetic oligonucleotide-based therapeutics.
